Product Description
|
Product Description: |
Inorganic pyrophosphatase (PPase) catalyzes the hydrolysis of inorganic pyrophosphate to form orthophosphate: P2O7-4 +H2O+PPase → 2HPO4-2). The enzyme requires a divalent metal cation, with Mg2+ conferring the highest activity. This product exerts its effect at 16-37℃, showing the strongest effect at 25℃. The Inactivation temperature is above 65℃. |
|
Applications: |
l High yield RNA synthesis by in vitro transcription. l DNA polymerization reactions: preventing accumulation of pyrophosphate. l Removal of contaminant PPi in reagents used for SNP genotyping by methods based on the detection of pyrophosphate. |
|
Source: |
An E. coli strain carrying a plasmid encoding a pyrophosphatase from the extreme thermophile Thermococcus litoralis. |

Definition of Activity Unit: |
One unit is the amount of enzyme that will generate 1 μmol of phosphate per minute from inorganic pyrophosphate under standard reaction conditions (a 10 minute reaction at 75°C in 100 mM Tris-HCl [pH 8.0], 2 mM MgCl2, 2 mM PPi, reaction volume of 0.5 ml). |
|
Storage (Dilution) Buffer: |
20 mM Tris-HCl, 100 mM NaCl, 1 mM DTT, 0.1 mM EDTA, 50% Glycerol, pH 8.0. |
|
Inactivation or inhibition |
l Inhibitors: imidodiphosphate, α ω glycol disphosphates, methanedial diphosphate, 1,2-ethanedial diphosphate. · l Inactivation by heating is not complete, reliably removed by spin column or phenol/chloroform extraction. |
|
Storage Conditions: |
Store at –20°C up to 12 months. |
