Product Description/Introduction
Hematoxylin is usually used in combination with Eosin for staining. Hematoxylin is oxidized to the acid dye hematoxylin, which combines with aluminum ions to form a positively charged blue pigment that binds to the negatively charged DNA in cells. Hematoxylinis an alkaline dye which mainly makes the chromatin in the nucleus and the ribosome in the cytoplasm blue to purple blue,Eosin Y is an acidic cytoplasmic dye, which mainly makes the cytoplasm and extracellular matrix components red.
This product is low toxicity and environmental protection, which can ensure the bright red and blue color of pathological sections are sharp and firm, with clear nucleoplasmic contrast, and will not fade in long-term storage. It can make the staining effect of the first to the 2500 sections basically consistent, and realize high resolution constant staining, which is conducive to the standardization and unification of HE staining and pathological diagnosis.G1076-2 hematoxylin dye solution is alcohol-soluble, pH 4.2~4.5.
Storage and Shipping Conditions
Store at room temperature (15-30℃) and valid for 18 months. Once opened should be used within 4 months. Keep the cap tightly after use to avoid volatilization or deterioration of the reagents.

Product protocol/procedures
1. Pre-treatment of tissue sections or cell crawls:
a) Dewaxing paraffin sections to water: environmentally friendly dewaxing solution I, II (G1028 recommended) or xylene I, IIfor15 min each; Anhydrous ethanol I, II, IIIfor5 min each; 75% ethanolfor2 min; Wash with water for 1 min.
b) Frozen sections store at -20℃, leave to rewarm at room temperature until the tissuesare slightly dried, and then fix with methanol (self-provided) for 1 min; Fresh frozen sectionsleave at room temperature for a few minutes, and thenstaining after OCT embedding agent completely dried to prevent desquamation.
c) Cell crawls fix with 4% paraformaldehyde for 5-10 min and wash with water for 1 min.
2. The sectionsare treated with high resolution constant dye pretreatment solution for 1 min.
3. Dye with hematoxylin solution for 3 min and wash with water for 1 min.
4. Differentiate with constant dye differentiation solution for 3-5 s, washwith waterfor 20 s.
5. Constant dye reblue solution for 1 min and wash with water for 20 s.
6. 95% ethanol for 60 s.
7. Stain with eosin Y dye solution for 15-30 s.
8. Anhydrous ethanol I, II, IIIfor2 min each.
9. N-butanolfor2 min, xylene transparentfor2 min.
10. Neutral gum seal piece.
Note
1. Read the instructions carefully before use, and use within the life of the product.
2. The product shall be stored according to the storage temperature. If the storage temperature is lower than 15℃,alum crystals may be precipitated from the hematoxylin staining solution, which generally does not affect the product quality, but may reduce the number of stained sections. If the storage temperature is higher than 30℃, the oxidation of hematoxylin will be accelerated, resulting in the decrease of dyeing ability, and the volatilization of eosin dyeing solution will be accelerated.
3. The optimal dyeing time of hematoxylin and eosin should be tested according to user samples.
4. Please wear lab coat and disposable gloves during operation. Prohibit inhalation by mouth or nose, and avoid direct contact with skin and mucous membrane. In case of contact with sensitive parts, rinse with plenty of water in time.
