Description
This product AB-PAS Stain is a combination of Alcian Blue (AB) and PAS dye solution. Alcian, also known as alcian blue, is a kind of copper peptide cyanine dye, which is alkaline and belongs to cationic dye. The dye can combine with acid groups to color tissues with anionic groups (such as carboxyl group). Alcian blue is a specific dye to display acidic mucus substances, which makes acidic mucopolysaccharides in the cytoplasm appear blue, while neutral mucins do not stain. PAS dye solution is used for glycogen PAS staining, also known as periodate Schiff stain, which is mainly used to detect glycogen and polysaccharide substances in tissues in histomorphology. The main principle is to oxidize the hydroxyl groups on the two adjacent carbon atoms in the carbohydrate materials to aldehyde groups through the oxidant periodic acid, and then react with the aldehyde groups with Schiff reagent to produce purple red substances. The acid glycoprotein and neutral glycoprotein can be distinguished by the combination of Alcian blue and PAS dye.
This product is a set combination. AB-PAS staining solution A is Schiff reagent, AB-PAS staining solution B is 0.5% periodate solution, and AB-PAS staining solution C is Alcian blue, with a concentration of 1% and a pH value of about 2.5. After the sample is dyed with this product, acidic mucus substances are blue, glycogen and neutral mucus substances are red, and mixed mucus substances are blue purple or purple blue. Generally speaking, in normal gastrointestinal mucus, the surface epithelium of gastric mucosa, pyloric gland, duodenal gland, etc. are mainly red neutral mucus substances. The goblet cells and intestinal glands of the small intestine and large intestine mainly secrete acidic mucus.
Storage and Handling Conditions
Wet ice transportation; The whole set of reagents shall be stored at 2-8℃, and the validity period shall be 12 months. Among them, AB-PAS staining solution B and AB-PAS staining solution C can also be stored at room temperature.

Assay Protocol
Preparation before experiment
Prepare xylene, gradient ethanol, absolute ethanol, neutral gum, etc.
usage method
1. Dewaxing of paraffin slices to water: the slices are dewaxed by xylene for 10 min in sequence, dewaxed by fresh xylene for 10 min, absolute ethanol for 5 min, fresh absolute ethanol for 5 min, 90% ethanol for 5 min, 75% ethanol for 5 min, and washed with tap water.
2. Slices into AB-PAS staining solution C for 8-10 min, and wash the slides with tap water until the running water is colorless.
3. The slices are immersed in AB-PAS staining solution B for 10-15 min, and then soaked in pure water for 3 times, about 10 s each.
4. The slices are put into AB-PAS staining solution A (recovered to room temperature in advance), covered and dipped in dark for 25-30 min, and washed with running water for 5 min.
5. Slice into three cylinders of anhydrous ethanol for dehydration, 5 min each time; After the xylene is transparent for 5 min, replace it with fresh xylene for another 5 min, and then mounted with neutral gum.
Note
1. AB-PAS staining solution B can be reused. The staining solution is transparent. If it appears obvious yellow, has impurities, and the staining of tissues is too light, it needs to be replaced with a new staining solution.
2. AB-PAS staining solution A needs to be stored at 4℃, taken out and recovered to room temperature before use. When the color of the solution is obviously red, it is recommended to replace it with a new staining solution.
3. Each set of staining solution (20 mL size) can be used to stain approximately 80 sections. Replace the staining solution with a new one when the tissue or cell coloring is significantly lighter or abnormal.
4. Please wear lab clothes and disposable gloves during operation.
