Product Description/Introduction

 

miRNA First Strand cDNA Synthesis Kit (Tailing Reaction) contains all the reagents required for miRNA first strand cDNA synthesis. The kit uses total RNA or miRNA as template, and Poly(A) Polymerase tailing reaction and reverse transcription reaction are synchronized to synthesize various miRNA first strand cDNAs with high efficiency. The kit also provides U6 Forward Primer and Universal Reverse Primer for subsequent amplification (applicable to human, rat and mouse samples), and the synthesized single-stranded cDNA product only needs to be designed with forward PCR primer for subsequent PCR or qPCR.

Assay Protocol / Procedures

 

1. Recommended reaction system preparation (Recommended reaction system preparation on ice)

Add to 20 μL

a: the recommended total RNA is 2 ug (no more than 5 μg), and the recommended miRNA is 200 ng.

2. Gently mix and centrifuge.

3. Perform poly(A) polymerize and reverse transcription using the recommened thermal conditions below:

4. The cDNA reaction solution obtained is recommended for gradient dilution before being used as a template for subsequent fluorescence quantitative PCR.

 

Note

 

1. Care was taken to avoid RNase contamination during the operation.

2. The reverse transcription products can be stored at -20℃ for a short period. If long-term storage is required, it is recommended to store at -80℃ after packing and avoid repeated freeze-thaw cycles.

3. The kit contains universal downstream primers for quantitative PCR, and the upstream primers are miRNA-specific primers, designed according to the target miRNA, and in general the upstream primers are the target miRNA sequences that change U to T.
